Combination of a monoclonal antibody, **3F8** (in murine or humanized form, targets disialoganglioside GD2 on tumor cells), and **sargramostim** (a recombinant human granulocyte-macrophage colony-stimulating factor, GM-CSF). This drug regimen is used as an immunotherapy, primarily in high-risk neuroblastoma and also in trials for osteosarcoma. 3F8 exerts antitumor effects primarily via antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) against GD2-expressing tumor cells. Sargramostim enhances the immune effector function of granulocytes and monocytes, augmenting the ADCC efficacy of 3F8. The combination aims to eradicate minimal residual disease by boosting both direct tumor killing and immune-mediated destruction. Developed and studied predominantly at Memorial Sloan Kettering Cancer Center.
