MAb 52-29.1 is a preclinical-stage monoclonal antibody developed by MedAbome that targets CD166 (activated leukocyte cell adhesion molecule, or ALCAM). Discovered using MedAbome's proprietary live-cell immunization (LC-I) and live-cell high-throughput screening (LC-HTS) platforms, MAb 52-29.1 selectively binds to a tumor-restricted conformational epitope of CD166 with high affinity (KD of approximately 1.1 nM) and minimal cross-reactivity with normal tissues. MedAbome is developing MAb 52-29.1 as a versatile therapeutic backbone across multiple modalities, including antibody-drug conjugates (ADCs), radiopharmaceutical drug conjugates (RDCs), and in vivo CAR-T/NK cell therapies. In preclinical studies, an antibody-drug conjugate version (MAb52-29.1-ADC) conjugated to monomethyl auristatin E (MMAE) demonstrated potent in vitro cytotoxicity and robust in vivo anti-tumor efficacy in xenograft models of solid tumors, including triple-negative breast cancer, non-small cell lung cancer, and gastric cancer.
