MAb 52-4.2 (also referred to as MAb52-4.2) is a preclinical-stage monoclonal antibody developed by MedAbome that targets Solute carrier family 3 member 2 (SLC3A2, also known as CD98hc or 4F2hc). Discovered using MedAbome's proprietary live-cell immunization (LC-I) and live-cell high-throughput screening (LC-HTS) platforms, MAb 52-4.2 selectively binds to a tumor-associated conformational epitope of SLC3A2 with high affinity, showing minimal cross-reactivity with normal tissues. MedAbome is optimizing MAb 52-4.2 for various therapeutic modalities, including antibody-drug conjugates (ADCs) such as MAb52-4.2-ADC (conjugated to monomethyl auristatin E [MMAE] via an MC-Vc-PAB linker), radionuclide drug conjugates (RDCs), and in vivo CAR-T/NK cell therapies. It is being investigated for the treatment of solid tumors (including triple-negative breast cancer, non-small cell lung cancer, gastric cancer, colorectal cancer, and pancreatic cancer) and hematologic malignancies.
