MAb 520C9 is a murine IgG1 monoclonal antibody that specifically targets the HER-2/neu (c-erbB2) oncoprotein, which is overexpressed in approximately 30% of breast cancers. Developed by Medarex, this antibody has been primarily utilized as a targeting component in the construction of bispecific antibodies, most notably MDX-H210. MDX-H210 is a bispecific construct created by chemically cross-linking the F(ab') fragment of MAb 520C9 with the F(ab') fragment of the humanized anti-CD64 antibody H22. The therapeutic mechanism involves linking HER-2/neu-expressing tumor cells to FcγRI-positive immune effector cells (such as monocytes, macrophages, and neutrophils) to facilitate antibody-dependent cell-mediated cytotoxicity (ADCC). MAb 520C9 is produced by the HB8696 hybridoma cell line and has been investigated in clinical trials as part of bispecific therapy for advanced breast cancer and other HER-2/neu-positive malignancies.
