Mab AR9.6 (also known as AR9.6 or huAR9.6) is a humanized monoclonal antibody developed by Quest PharmaTech, originally licensed from the University of Nebraska Medical Center. It targets a novel tumor-associated marker, truncated O-glycan-bearing MUC16 (mucin-16), specifically binding to its SEA (Sea urchin sperm, Enterokinase, Agrin) domains. By binding to MUC16, Mab AR9.6 blocks the interaction of MUC16 with ErbB (EGF) receptors and alpha4beta1 integrin on the cancer cell surface, thereby inhibiting downstream oncogenic signaling pathways, including AKT and GSK3beta phosphorylation. This leads to reduced cell proliferation, tumor growth, and metastasis. Mab AR9.6 is in preclinical development for the treatment of pancreatic cancer, ovarian cancer, and breast cancer, and is also being investigated as a theranostic agent for immuno-PET imaging, fluorescence-guided surgery, and radioimmunotherapy.
