Overview

MDX-33 (also known as Mab h22) is a humanized monoclonal antibody that targets the high-affinity immunoglobulin gamma Fc receptor I (CD64) on macrophages. By binding to CD64, it acts as an antagonist and inhibits the activity of monocytes and macrophages, thereby reducing their ability to destroy healthy platelets. This mechanism was investigated for potential therapeutic benefit in immune-mediated hematological disorders such as idiopathic thrombocytopenic purpura (ITP), autoimmune hemolytic anemia, and thrombocytopenia. The drug was developed by Medarex and Bristol Myers Squibb but has been discontinued after early-phase clinical trials[1][2][4].
