mAb W9 is a human monoclonal antibody that specifically targets an extracellular epitope of the heat shock protein Grp94 (glucose-regulated protein of 94 kDa), a member of the HSP90 family. Grp94 is overexpressed on the surface of various malignant cells, including differentiated pancreatic adenocarcinoma cells and pancreatic cancer initiating cells (CICs), which are associated with disease recurrence and chemoresistance. By binding to cell-surface Grp94, mAb W9 inhibits tumor cell growth and induces apoptosis. Its mechanism of action involves the downregulation of key signaling pathways, including RAS/Raf/MEK/ERK and PI3K/AKT. Preclinical studies have shown that mAb W9 exhibits significant antiproliferative activity in pancreatic cancer models, particularly when combined with hedgehog signaling inhibitors like cyclopamine and chemotherapy such as 5-fluorouracil (5-FU).
