MAb007 is a recombinant monoclonal IgG1 antibody characterized by an unpaired cysteine residue located in its CDR3 variable region. This free cysteine is prone to cysteinylation, a post-translational modification that introduces heterogeneity and adversely affects the antibody's stability and biological activity. Studies have shown that cysteinylation can increase the rate of MAb007 aggregation and decrease the melting temperature of its Fab region. A mild redox step has been identified as an effective method to remove cysteinylation, thereby improving the antibody's homogeneity, stability, and biological activity.
