**mAb104-DM1** is a preclinical HER2-targeted antibody-drug conjugate composed of the murine IgG1 monoclonal antibody mAb104 linked through an SMCC linker to the maytansinoid microtubule inhibitor DM1. mAb104 recognizes a tumour-selective conformational epitope on human epidermal growth factor receptor 2 that is exposed on tumour cells while showing minimal normal-tissue binding. Following HER2 binding and tumour-cell internalisation, intracellular release or processing of the DM1 conjugate disrupts microtubule function and inhibits cell division. The ADC was prepared by Levena Biopharma with a drug-to-antibody ratio of approximately 3.4 to 3.7 and has shown dose-dependent cytotoxicity and antitumour activity in preclinical HER2-expressing cancer models.
