MAb173 is an experimental monoclonal antibody that targets the AXL receptor tyrosine kinase. AXL is a member of the TAM (Tyro3, Axl, Mer) family and is frequently overexpressed in various cancers, where it contributes to tumor progression, epithelial-to-mesenchymal transition (EMT), and resistance to targeted therapies, particularly EGFR inhibitors like cetuximab. MAb173 works by binding to the extracellular domain of AXL, leading to receptor internalization and degradation, thereby inhibiting downstream signaling pathways such as MAPK/ERK and PI3K/Akt. Preclinical research has demonstrated its potential in sensitizing cetuximab-resistant non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma (HNSCC) to therapy by disrupting the positive feedback loop between EGFR and AXL.
