Overview

The name **mAb2** refers to two distinct experimental monoclonal antibodies described in the provided context. 1. **csNPM1-targeting mAb2**: This is a mouse IgG2a monoclonal antibody developed to target cell surface nucleophosmin (csNPM1). While NPM1 is typically a nucleolar protein, it is aberrantly expressed on the surface of various cancer cells, particularly in Acute Myeloid Leukemia (AML), where it serves as a tumor-specific antigen. This mAb2 binds to csNPM1 in a mutation-agnostic manner, targeting both wild-type and mutant (NPM1c) forms. Its primary mechanism of action involves opsonizing cancer cells to induce antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Preclinical studies demonstrate that it effectively targets leukemic stem cells (LSCs) and blasts while sparing healthy hematopoietic stem cells, and shows efficacy in syngeneic models of prostate and colorectal cancer. 2. **GIPR-targeting mAb2**: This is a full antagonistic monoclonal antibody directed against the glucose-dependent insulinotropic polypeptide receptor (GIPR). It binds to the GIPR C-terminal stalk region, acting as a molecular mimic of the ligand peptide to lock the receptor in an auto-inhibited state. It has been studied for its potential to improve metabolic profiles and reduce body weight gain in models of diet-induced obesity.
