mAb3 is a chimeric monoclonal antibody designed to target the programmed cell death-1 (PD-1) receptor. Developed by Amgen Research, it features an anti-human PD-1 variable region fused to a mouse IgG1 constant region, a configuration specifically optimized for evaluation in humanized mouse models. mAb3 functions as a checkpoint inhibitor by binding to PD-1 on T cells and blocking its interaction with the ligands PD-L1 and PD-L2. This blockade prevents the inhibitory signaling that typically suppresses T-cell activation, thereby enhancing the immune system's ability to recognize and eradicate tumor cells. In preclinical research, mAb3 has been utilized both as a monotherapy control and as the foundational targeting component for bifunctional fusion proteins (immunocytokines), such as those incorporating engineered interleukin-21 (IL-21) muteins to deliver localized cytokine signaling to PD-1-expressing T cells.
