Overview

mAb428.2 is a function-blocking monoclonal antibody targeting the tumor extracellular matrix protein fibulin-3 (EFEMP1). Fibulin-3 is a biomarker for malignant mesothelioma (MM) and is involved in tumor progression by activating canonical NFkB signaling, which in turn regulates proteases like MMP9 and MMP13, and proinvasive proteins such as TNC and MLCK. mAb428.2 binds to a specific epitope of fibulin-3 required for NFkB activation, thereby inhibiting downstream signaling, reducing tumor cell viability, and enhancing the cytotoxicity of chemotherapy agents like cisplatin. In preclinical models of malignant mesothelioma, the antibody has demonstrated significant reduction in tumor burden and extension of overall survival.
