MAb51-31-ADC is a first-in-class antibody-drug conjugate (ADC) developed by MedAbome, Inc., targeting a tumor-restricted conformational epitope of Integrin α/β (ITGα/β). The ADC consists of a humanized monoclonal antibody (MAb51-31) conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a protease-cleavable MC-Vc-PAB linker, with a drug-to-antibody ratio (DAR) of 4. MAb51-31-ADC is designed to selectively target ITGα/β, a heterodimeric transmembrane glycoprotein overexpressed in various solid tumors that promotes cancer progression and metastasis. In preclinical studies, the agent demonstrated potent antiproliferative effects and complete tumor regression in xenograft models of triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and gastric cancer (GC), while showing no cross-reactivity with normal tissues.
