MAb52-29.1-ADC is a first-in-class antibody-drug conjugate (ADC) targeting Activated Leukocyte Cell Adhesion Molecule (ALCAM/CD166), a glycoprotein overexpressed in various solid and hematologic malignancies. Developed by MedAbome, Inc., the ADC consists of a monoclonal antibody (MAb52-29.1) that recognizes a tumor-restricted conformational epitope of ALCAM, conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a protease-cleavable MC-Vc-PAB linker with a drug-to-antibody ratio (DAR) of 4. The antibody component exhibits high binding affinity and efficient internalization. In preclinical studies, MAb52-29.1-ADC demonstrated potent anti-tumor efficacy in xenograft models of triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and gastric cancer, with a favorable safety profile in preliminary toxicology assessments.
