mAb9.2.27 is a monoclonal antibody that targets the chondroitin sulfate proteoglycan 4 (CSPG4), also known as neuron-glial antigen 2 (NG2). CSPG4 is a cell surface proteoglycan highly expressed in various aggressive malignancies, including melanoma and glioblastoma multiforme (GBM), where it plays a critical role in tumor cell proliferation, migration, and angiogenesis. In preclinical studies, mAb9.2.27 has demonstrated the ability to inhibit tumor growth and modulate the immune microenvironment. Specifically, in glioblastoma models, its intracranial administration in combination with activated natural killer (NK) cells has been shown to shift microglia from a pro-tumorigenic M2 phenotype to a cytotoxic M1-like phenotype, facilitating a Th1 immune response and increasing tumor infiltration by CD4+ and CD8+ T cells. This combined approach aims to eradicate established tumors and prolong survival by enhancing local immune-mediated cytotoxicity.
