mAb_Ent is a therapeutic monoclonal antibody (IgG1) being developed by Immunethep for the treatment of infections caused by Enterobacteriaceae, specifically Escherichia coli and Klebsiella pneumoniae. The antibody targets extracellular bacterial glyceraldehyde-3-phosphate dehydrogenase (bGAPDH), a conserved virulence factor excreted by these pathogens to suppress the host's immune system. In the extracellular environment, bGAPDH normally binds to toll-like receptor 2 (TLR2) on B lymphocytes, triggering rapid production of the anti-inflammatory cytokine interleukin-10 (IL-10) and causing systemic immunosuppression. By neutralizing bGAPDH, mAb_Ent prevents this immunosuppressive cascade, restoring the host's natural immune response to control and eliminate the bacterial infection without inducing antimicrobial resistance.
