MABS-01 is a preclinical-stage antibody-drug conjugate (ADC) being developed by Manhattan BioSolutions for the treatment of pediatric and adult cancers. The therapeutic consists of a monoclonal antibody conjugated to a topoisomerase 1 inhibitor (TOP1i) payload. MABS-01 is designed to selectively target tumor cells and deliver its cytotoxic payload intracellularly, where it inhibits the DNA topoisomerase 1 enzyme. This inhibition leads to the stabilization of cleavage complexes, resulting in DNA strand breaks and subsequent apoptosis of the cancer cell. The program is currently undergoing preclinical evaluation to support potential future clinical development.
