Overview

MAC28 is a novel small molecule inhibitor targeting myeloid differentiation factor 2 (MD-2), a co-receptor of toll-like receptor 4 (TLR4) involved in pro-inflammatory signaling. MAC28 binds directly to MD-2, blocking its interaction with lipopolysaccharide and attenuating downstream TLR4-driven pathways. Experimental studies show that MAC28 suppresses inflammation and oxidative stress by downregulating MD2/TLR4 signaling and inflammatory cytokines (such as TNF-α and IL-6), and restores mitochondrial and tissue function. It has demonstrated efficacy in preclinical models for reducing left ventricular dysfunction in prediabetic rats, attenuating neurodegeneration and improving cognitive function, and reducing inflammation and restoring cell differentiation in dental pulp cell models. MAC28 is reported to be a synthetic derivative related to curcumin and was provided by Wenzhou Medical University for research purposes[1][2][3][4].
