MACH-1 Seasonal Flu Vaccine is a preclinical nucleic acid vaccine candidate being developed by Orlance for the prevention of seasonal influenza. The vaccine utilizes the proprietary MACH-1 (Micro-particle Accelerated Cellular High-efficiency) platform, which employs a needle-free "gene gun" to deliver DNA-coated gold microparticles directly into the epidermal layer of the skin. This delivery method targets the immune-rich environment of the epidermis, specifically transfecting keratinocytes and professional antigen-presenting cells such as Langerhans cells. Once inside the cells, the DNA plasmids encode influenza antigens—typically Hemagglutinin (HA) and potentially Neuraminidase (NA)—which are then expressed and presented to the immune system. This process triggers a robust and balanced immune response, including the production of neutralizing antibodies and the activation of T-cell mediated immunity. The MACH-1 platform is designed to provide more effective protection than traditional egg-based vaccines and offers advantages in terms of stability and ease of administration.
