Drug intelligence / Profile preview
Macs@EcN IL-15 is a preclinical-stage, macrophage-encapsulated engineered bacterial therapy developed by Nanjing University for the treatment of bladder cancer. The therapeutic agent consists of a recombinant *Escherichia coli* Nissle 1917 (EcN) strain engineered to express and secrete an interleukin-15 (IL-15) and IL-15 receptor alpha (IL-15Rα) fusion protein (EcN IL-15). This engineered strain is encapsulated within bone marrow-derived macrophages (BMDMs), which leverage their natural tumor-specific tropism and adhesion capabilities to target bladder tumors. Upon intravesical administration, the macrophages deliver the bacteria directly to the tumor microenvironment, where the sustained release of the IL-15/IL-15Rα complex activates and expands T cells and natural killer (NK) cells, promoting localized, durable antitumor immune responses.
