Madecassoside is a triterpenoid saponin and a major phytoactive constituent of Centella asiatica (Gotu kola), a medicinal plant widely used in traditional Chinese, Ayurvedic, and African medicine. It exhibits broad pharmacological activities including anti-inflammatory, antioxidant, wound-healing, neuroprotective, and skin-protective effects[1][3][5]. Mechanistically, madecassoside suppresses pro-inflammatory cytokine production by inhibiting ERK, p38 MAPK, and NF-κB signaling pathways[3][4]. It also promotes collagen synthesis and angiogenesis in the skin while reducing oxidative stress markers such as malondialdehyde (MDA) and nitric oxide (NO)[6]. Madecassoside has demonstrated efficacy in facilitating wound closure in animal models and improving insulin sensitivity. Additionally, it inhibits melanin synthesis by blocking ultraviolet-induced inflammation through downregulation of protease-activated receptor-2 signaling[8]. Its neuroprotective properties are attributed to its ability to reduce neuronal apoptosis via modulation of Bcl-2/Bax expression ratios[4].
