MAG-007 is a third-generation, small molecule inhibitor of Bruton's tyrosine kinase (BTK) developed by Miragene. As a non-covalent and reversible inhibitor, MAG-007 is designed to overcome the limitations of first-generation covalent BTK inhibitors, such as resistance mediated by the C481S mutation and off-target safety concerns. A key differentiating feature of MAG-007 is its ability to penetrate the blood-brain barrier, which may allow for the treatment of neuroinflammatory and autoimmune conditions of the central nervous system. The drug is currently being developed for the treatment of chronic spontaneous urticaria (CSU) and has completed Phase 1 clinical testing, positioning it as Phase 2-ready with potential for once-daily oral dosing.
