MAG-Tn3 is a fully synthetic glycopeptide therapeutic cancer vaccine designed to induce an immune response against the Tn antigen, a tumor-associated carbohydrate antigen highly expressed in many carcinomas, especially breast carcinoma. The structure of MAG-Tn3 consists of three consecutive Tn moieties O-linked to a CD4+ T-cell epitope derived from tetanus toxoid (TT830–844), forming a dendrimeric peptide backbone. This design enables robust activation of CD4+ T-cells and production of anti-Tn antibodies. The vaccine is formulated with the AS15 immunostimulant (containing CpG7909, MPL, and QS21) to enhance both innate and adaptive immune responses. In phase I clinical trials for high-risk localized breast cancer patients, immunization with MAG-Tn3/AS15 induced strong Tn-specific IgM and IgG antibody responses capable of recognizing and killing Tn-expressing tumor cells via complement-dependent cytotoxicity[1][2][6][8].
