MAGE-1.A2 peptide vaccine is an investigational peptide-based therapeutic cancer vaccine developed by Cliniques Universitaires Saint-Luc (Université Catholique de Louvain) in Brussels, Belgium. The vaccine consists of a synthetic peptide corresponding to the HLA-A2-restricted epitope of the Melanoma-Associated Gene 1 (MAGE-1/MAGEA1) protein. It is designed to stimulate cytolytic T-lymphocyte (CTL) responses against melanoma cells expressing the MAGE-1 antigen. In clinical development, it was evaluated as part of an eight-peptide cocktail (including MAGE-3.A2, MAGE-4.A2, MAGE-10.A2, MAGE-C2.A2, NA17.A2, Tyrosinase.A2, and NY-ESO-1.A2) administered with or without adjuvants such as Montanide ISA 51 and IMP321 (eftilagimod alpha) in a Phase 1/2 trial (NCT00365937) for disease-free, HLA-A2-positive cutaneous melanoma patients at high risk of relapse. The trial was terminated due to regulatory requirements from the peptide supplier.
