MAGE-3 antigen specific T cells are a form of adoptive cell therapy in which autologous or allogeneic cytotoxic CD8+ T cells are genetically engineered to express a high-affinity, tumor-specific T cell receptor (TCR) that recognizes the HLA-restricted epitope of the melanoma-associated antigen 3 (MAGE-A3). MAGE-A3 is a cancer-testis antigen highly expressed in various tumors, including melanoma and non-small cell lung cancer, but not in normal tissues except for testis. These engineered T cells are designed to target and kill tumor cells expressing MAGE-A3 by recognizing peptide fragments presented on HLA molecules. The approach aims to enhance anti-tumor immunity with reduced risk of off-target toxicity due to the restricted expression profile of MAGE antigens[1]. This strategy is under investigation primarily for solid tumors such as melanoma and non-small cell lung cancer.
