MAGE-3.1 is a recombinant antigen derived from the Melanoma-associated antigen 3 (MAGE-A3), a member of the cancer-testis antigen family. MAGE-A3 is an ideal target for cancer immunotherapy as it is highly expressed in various malignancies, including melanoma, but is absent in normal adult tissues except for the testis and placenta. Developed and evaluated by the University of Southern California (USC), MAGE-3.1 is designed to stimulate the immune system, specifically inducing a cytotoxic T-lymphocyte (CTL) response against tumor cells that express the MAGE-A3 protein. In clinical trial settings, it is often administered as part of a multi-antigen vaccine regimen alongside other melanoma-associated antigens such as gp100 and tyrosinase. To enhance its immunogenicity and overcome tumor-induced immune suppression, it is typically formulated with adjuvants like Montanide ISA 51 and the Toll-like receptor 9 (TLR9) agonist CpG 7909 (agatolimod sodium).
