MAGE-3.A1 + NA17.A2 + GM-CT-01 is an experimental combination immunotherapy regimen developed for the treatment of advanced metastatic melanoma. The therapy consists of two tumor-specific antigenic peptides, MAGE-3.A1 (derived from the MAGE-A3 protein) and NA17.A2 (derived from the N-acetylglucosaminyltransferase V protein), which are administered as a therapeutic vaccine to stimulate cytotoxic T lymphocyte (CTL) responses against melanoma cells. This vaccine is used in conjunction with GM-CT-01 (davnatide), a galactomannan-based galectin-3 inhibitor. Galectin-3 is a protein often secreted by tumors to induce anergy in tumor-infiltrating lymphocytes (TILs), thereby creating an immunosuppressive microenvironment. By inhibiting galectin-3, GM-CT-01 is intended to restore T-cell activity and enhance the efficacy of the immune response generated by the peptide vaccination. This combination was evaluated in Phase I/II clinical trials led by Cliniques universitaires Saint-Luc and the Université Catholique de Louvain in collaboration with the Ludwig Institute for Cancer Research and Galectin Therapeutics.
