**MAGE-3.A1 peptide** is a synthetic peptide derived from the tumor antigen *MAGE-3* (also known as *MAGE-A3*), designed for use as a cancer vaccine, especially in metastatic melanoma. The peptide is HLA-A1 restricted, meaning it is presented by HLA-A1 molecules to cytotoxic T lymphocytes (CTLs). Its primary mechanism is as an immunotherapeutic agent, aiming to induce a CTL response against tumor cells expressing MAGE-3. Clinical studies have shown that subcutaneous and dermal injection, or dendritic cell (DC) vaccine formulations using the peptide, can result in partial or complete tumor regression in a subset of advanced melanoma patients, sometimes with durable responses lasting years[1][2][3][4]. The peptide is strictly tumor-specific, as MAGE-3 is expressed in various tumors but not in normal tissues except immune-privileged male germ cells[4][5]. The mechanism of action involves stimulating CD8+ cytotoxic T lymphocyte precursors (CTLp) that recognize the peptide when presented by HLA-A1 on tumor cells, potentially leading to in situ tumor cell killing[2][4]. MAGE-A3 is also a target in other cancers, including NSCLC, bladder, esophageal, and breast cancers[5].
