MAGE-3.A2 is a melanoma-associated peptide vaccine component developed by the Ludwig Institute for Cancer Research. It consists of a specific HLA-A2-restricted cytotoxic T-lymphocyte (CTL) epitope (sequence FLWGPRALV) derived from the MAGE-3 (also known as MAGE-A3) tumor antigen. MAGE-3 is a cancer-germline (cancer-testis) antigen that is expressed in various malignancies, including melanoma, but is absent in normal adult tissues except for the testis, making it a highly specific target for immunotherapy. The peptide is designed to be presented by HLA-A2 MHC class I molecules on the surface of cells, thereby activating CD8+ cytotoxic T lymphocytes to recognize and destroy MAGE-3-expressing tumor cells. In clinical trials, it has been evaluated as part of multi-peptide vaccine formulations, often in combination with immunological adjuvants like CpG 7909, for the treatment of HLA-A2 positive patients with metastatic melanoma.
