MAGE-3.A2 peptide vaccine is a cancer immunotherapy peptide vaccine developed and studied by Cliniques Universitaires Saint-Luc (Université Catholique de Louvain) in Belgium. It consists of a synthetic peptide derived from the Melanoma Antigen Gene-3 (MAGE-3) protein (sequence: FLWGPRALV) that binds to the HLA-A2 MHC class I molecule. The vaccine is designed to stimulate an HLA-A2-restricted cytolytic T-lymphocyte (CTL) immune response against MAGE-3-expressing tumor cells. It was evaluated in a Phase 2 clinical trial (NCT00365937) as part of an eight-peptide mixture (including MAGE-1.A2, MAGE-3.A2, MAGE-4.A2, MAGE-10.A2, MAGE-C2.A2, NA17.A2, Tyrosinase.A2, and NY-ESO-1.A2) administered with or without adjuvants like Montanide ISA 51 and IMP321 in patients with high-risk cutaneous melanoma. The development of the vaccine was terminated due to regulatory issues with the peptide supplier.
