Overview

MAGE-4.A2 is a melanoma-associated peptide antigen derived from the MAGE-A4 (Melanoma-Associated Antigen 4) cancer/testis protein. It is an HLA-A2-restricted epitope designed for use in cancer immunotherapy, specifically as a component of multi-peptide therapeutic vaccines. Developed by the Ludwig Institute for Cancer Research, it has been evaluated in Phase I/II clinical trials for patients with HLA-A2 positive metastatic melanoma. The vaccine typically combines MAGE-4.A2 with other melanoma-associated peptides and an immunological adjuvant, such as the TLR9 agonist CpG 7909, to induce a cytotoxic T-lymphocyte (CTL) response against tumor cells expressing the MAGE-A4 antigen. MAGE-A4 is a highly specific target as it is expressed in various malignancies but restricted to the testis and placenta in normal adult tissues.
