MAGE-4.A2 peptide vaccine is an experimental, research-stage multipeptide cancer vaccine developed by Cliniques Universitaires Saint-Luc (Université Catholique de Louvain) for the adjuvant treatment of HLA-A2-positive, disease-free cutaneous melanoma patients at high risk of relapse. The vaccine consists of a mixture of eight synthetic, melanoma-associated HLA-A2-restricted peptides, including MAGE-1.A2, MAGE-3.A2, MAGE-4.A2, MAGE-10.A2, MAGE-C2.A2, NA17.A2, Tyrosinase.A2, and NY-ESO-1.A2. These peptides function as tumor-associated antigens that, when presented by HLA-A2 molecules, stimulate cytolytic T-lymphocytes (CTLs) to recognize and destroy melanoma cells. In clinical evaluation (such as the Phase 2 trial NCT00365937), the vaccine was administered via intradermal and subcutaneous injections, either alone or in combination with adjuvants like Montanide ISA 51 and/or the LAG-3Ig fusion protein IMP321 (eftilagimod alpha) to enhance immune responses. The development of this vaccine was terminated due to regulatory requirements imposed by the peptide supplier.
