MAGE-A10 peptide is a synthetic peptide vaccine component derived from the Melanoma-associated antigen 10 (MAGE-A10), a member of the cancer-testis antigen (CTA) family. CTAs are highly attractive targets for cancer immunotherapy because their expression is typically restricted to tumor cells and immune-privileged sites such as the testis and placenta, minimizing off-target toxicity. The MAGE-A10 peptide, often representing the HLA-A*0201-restricted epitope with the sequence GLYDGMEHL, is designed to be presented by Major Histocompatibility Complex (MHC) class I molecules on the surface of antigen-presenting cells. This presentation triggers the activation and expansion of antigen-specific CD8+ cytotoxic T lymphocytes (CTLs), which can then recognize and lyse MAGE-A10-expressing malignant cells. In clinical trials, such as those conducted by the Ludwig Institute for Cancer Research, the peptide is frequently administered with adjuvants like Montanide ISA-51 and CpG oligonucleotides to enhance the magnitude and duration of the systemic immune response. It has been primarily investigated for the treatment of advanced melanoma and other MAGE-A10-expressing malignancies.
