Drug intelligence / Profile preview
MAGE-A3 + NY-ESO-1 refers to a combination immunotherapy approach or vaccine targeting two cancer-testis antigens, MAGE-A3 and NY-ESO-1. These antigens are highly expressed in various tumors but not in normal tissues except for immune-privileged sites like the testis and placenta. The combination is designed to stimulate the immune system—particularly cytotoxic T lymphocytes—to recognize and destroy tumor cells expressing these antigens. This strategy is being explored primarily as a therapeutic cancer vaccine or as part of adoptive cell therapy regimens, often using viral vectors (such as ChAdOx1 or Modified Vaccinia Ankara) to deliver the antigenic payloads, or dendritic cell-based vaccines pulsed with peptides from both antigens[5][4][2]. The main indications under investigation include non-small cell lung cancer (NSCLC), multiple myeloma, neuroblastoma, sarcoma, and other solid tumors[5][4][2]. Mechanistically, this approach aims to induce robust antigen-specific T-cell responses against tumor cells while sparing healthy tissue due to the restricted expression profile of these antigens[5][6].
