MAGE-A3 ASCI (Antigen-Specific Cancer Immunotherapeutic) is a cancer vaccine consisting of a recombinant MAGE-A3 protein combined with the AS15 immunostimulant adjuvant system. Developed by GSK, it targets the MAGE-A3 (Melanoma-Associated Antigen A3) protein, a tumor-specific antigen expressed in various cancers including melanoma, non-small cell lung cancer, and bladder cancer, but absent in normal tissues except for the testis and placenta. The vaccine is designed to induce a specific immune response against tumor cells expressing the MAGE-A3 antigen. While it showed promise in early-phase trials, including a Phase II study at the H. Lee Moffitt Cancer Center assessing its use with or without the Poly IC:LC adjuvant in stage IV melanoma patients, larger Phase III trials (such as the DERMA trial in melanoma and the MAGRIT trial in NSCLC) failed to meet their primary endpoints, leading to the discontinuation of its development for these indications.
