MAGE-A3 ASCI + Poly IC:LC is a combination immunotherapeutic regimen designed for cancer treatment, particularly melanoma and multiple myeloma. The therapy consists of a recombinant MAGE-A3 antigen-specific cancer immunotherapeutic (ASCI), which targets the tumor-associated antigen MAGE-A3, combined with the adjuvant Poly IC:LC (Hiltonol), a synthetic double-stranded RNA that acts as an agonist of Toll-like receptor 3 (TLR3). The combination aims to enhance anti-tumor immune responses by stimulating both innate and adaptive immunity. Clinical trials have evaluated its safety and immunogenicity in patients with melanoma and multiple myeloma, showing that it can induce robust vaccine-specific T-cell responses when used alongside other agents such as GM-CSF[1][2][4][5].
