MAGE-A3-ImmTAC (also known as IMC-A121) is a first-generation bispecific T-cell engager developed by Immunocore using its proprietary ImmTAC (Immune-mobilizing monoclonal TCRs Against Cancer) platform. The molecule is a recombinant fusion protein comprising a soluble, affinity-matured T-cell receptor (TCR) specific for a MAGE-A3 peptide (presented by HLA-A*01) fused to an anti-CD3 single-chain antibody fragment (scFv). Its mechanism of action involves the TCR domain binding to MAGE-A3-positive cancer cells, while the anti-CD3 domain recruits and activates polyclonal T cells to induce tumor cell lysis. MAGE-A3 is a cancer-testis antigen expressed in various malignancies, including melanoma and non-small cell lung cancer, but not in normal tissues. Clinical development of MAGE-A3-ImmTAC was terminated following early-phase trials.
