MAGE-A3 Trojan peptide is an investigational cancer vaccine component designed to stimulate a robust immune response against tumors expressing the Melanoma-associated antigen 3 (MAGE-A3). The vaccine utilizes "Trojan" technology, which involves synthetic long peptides containing both CD4+ and CD8+ T-cell epitopes connected by furin-cleavable linkers. This specific design allows the peptide to be internalized and processed by the cell's endogenous furin protease, facilitating the presentation of epitopes on both MHC Class I and Class II molecules. By enhancing antigen processing and presentation, the MAGE-A3 Trojan peptide aims to overcome the immune defects often observed in patients with advanced cancers. It has been primarily evaluated in clinical trials for the treatment of MAGE-A3-positive Squamous Cell Carcinoma of the Head and Neck (SCCHN), typically administered in combination with adjuvants such as GM-CSF and Montanide ISA 51.
