MAGE-A3 vaccine is an investigational cancer immunotherapy designed to induce an immune response against tumor cells expressing the melanoma-associated antigen A3 (MAGE-A3), a cancer-testis antigen found in various malignancies including melanoma, lung, breast, brain, and ovarian cancers. Several formats have been developed: - Recombinant protein vaccines (often combined with adjuvants such as AS15 or AS02B) aim to stimulate both humoral and cellular immunity. - Viral vector-based vaccines use either non-replicating adenovirus vectors (Ad-MAGEA3) or oncolytic viruses (MG1-MAGEA3) engineered to express the MAGE-A3 antigen. These approaches prime and boost CD8+ T-cell responses against tumors. The primary mechanism involves presenting the MAGE-A3 antigen to the immune system to elicit cytotoxic T lymphocyte responses that target and kill tumor cells expressing this antigen[1][2][4][5][6][8]. GlaxoSmithKline led large clinical trials of a recombinant protein-based version for melanoma and non-small cell lung cancer; however, pivotal phase 3 studies did not meet their primary endpoints[6]. Research continues into improved formulations and delivery methods.
