MAGE-A4 CAR-T cells are a form of autologous chimeric antigen receptor (CAR) T cell therapy engineered to target the melanoma-associated antigen A4 (MAGE-A4), a cancer-testis antigen expressed in various solid tumors. These therapies use a patient’s own T lymphocytes that are genetically modified ex vivo to express a synthetic receptor—typically an scFv antibody fragment specific for the peptide-MHC complex of MAGE-A4 presented by HLA molecules. Some advanced constructs incorporate elements of the native T-cell receptor (TCR) machinery into the CAR structure to enhance tumor recognition and cytotoxicity while maintaining specificity. The approach aims to direct potent immune responses against solid tumors expressing MAGE-A4, such as synovial sarcoma, esophageal cancer, ovarian cancer, and others[6][7]. Preclinical studies have shown that these hybrid or next-generation designs can improve anti-tumor efficacy by increasing functional activity and persistence of infused T-cells[6][7].
