MAGE-A4-targeted CAR-T cells are an investigational autologous cell therapy being developed by Shanghai Tenth People's Hospital for the treatment of advanced solid tumors. The therapy involves genetically engineering a patient's own T cells to express a chimeric antigen receptor (CAR) that specifically recognizes Melanoma-Associated Antigen A4 (MAGE-A4). MAGE-A4 is a cancer-testis antigen that is highly expressed in various malignancies—such as synovial sarcoma, melanoma, and lung cancer—but has restricted expression in normal tissues, making it a suitable target for redirected T-cell therapy. Upon re-infusion, these CAR-T cells are designed to identify and eliminate MAGE-A4-expressing tumor cells. Clinical evaluation is currently focused on safety and preliminary efficacy in patients with MAGE-A4-positive solid tumors who have failed standard therapies.
