MAGE-A4-TCR is an autologous T-cell receptor (TCR) engineered T-cell therapy designed to target the Melanoma-Associated Antigen A4 (MAGE-A4), a cancer-testis antigen frequently expressed in various solid tumors. The TCR was discovered using Medigene's proprietary platform and was initially licensed to 2seventy bio before being acquired by Regeneron Pharmaceuticals in April 2024. The therapy works by modifying a patient's own T cells to express a specific TCR that recognizes MAGE-A4 peptides presented by HLA molecules on tumor cells, leading to targeted cell killing. It is currently in early-stage clinical development for the treatment of solid tumors, with specific trials being conducted in China through a partnership with JW Therapeutics.
