MAGE-C2 shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of the Melanoma-associated antigen C2 (MAGE-C2) gene. MAGE-C2 is a member of the cancer-testis antigen family that is frequently overexpressed in multiple myeloma (MM) and is associated with aggressive disease and chemotherapy resistance. Delivered via a lentiviral vector in preclinical models, the shRNA mediates stable gene silencing of MAGE-C2, which triggers the DNA damage response pathway involving ATM/ATR and CHK1/CHK2 kinases. This activation leads to the stabilization of the p53 protein, resulting in cell cycle arrest at the G0/G1 and G2/M transitions and the induction of apoptosis through the upregulation of pro-apoptotic genes such as BAX and BAK. This agent is primarily utilized in research to investigate MAGE-C2 as a potential target for myeloma-specific therapies.
