MAGE-C2.A2 is a peptide-based therapeutic cancer vaccine component derived from the MAGE-C2 (Melanoma Antigen Family C2) cancer-germline gene. It specifically consists of HLA-A2-restricted antigenic peptides, such as ALKDVEERV and LLFGLALIEV, designed to elicit cytotoxic T-lymphocyte (CTL) responses against melanoma cells. MAGE-C2 is a member of the cancer-testis antigen family, which is expressed in various malignancies, including approximately 40% of melanomas, but remains silent in normal somatic tissues except for the testis. Developed by the Ludwig Institute for Cancer Research, MAGE-C2.A2 has been investigated in Phase I/II clinical trials as part of a multi-peptide vaccine cocktail (often combined with other melanoma-associated peptides and the adjuvant CpG 7909) for the treatment of HLA-A2-positive patients with metastatic melanoma.
