MAGE3-TBHSP70 is a recombinant fusion protein vaccine candidate designed for cancer immunotherapy. It consists of a specific segment of the melanoma-associated antigen 3 (MAGE-3), typically amino acids 195-314, covalently linked to the C-terminus of Mycobacterium tuberculosis heat shock protein 70 (HSP70). In this construct, HSP70 serves as a potent molecular adjuvant and chaperone that enhances the recruitment, uptake, and processing of the MAGE-3 antigen by professional antigen-presenting cells. This linkage is essential for eliciting a robust, antigen-specific immune response, including the activation of cytotoxic T lymphocytes (CTLs) and the production of cytokines. Preclinical studies have demonstrated that MAGE3-TBHSP70 can induce significant tumor regression and prolong survival in animal models of MAGE-3-expressing malignancies, such as melanoma, hepatocellular carcinoma, and pulmonary carcinoma.
