MAGED1 small interfering RNA is an experimental RNA interference (RNAi) agent designed to silence the expression of the Melanoma Antigen Gene Family, Member D1 (MAGED1). MAGED1 is a multi-functional protein involved in cell cycle regulation and cell death, and it plays a critical role in the repair of DNA double-strand breaks via the homologous recombination repair (HRR) pathway. Suppression of MAGED1 leads to homologous recombination deficiency (HRD), a state that significantly increases the sensitivity of cancer cells to platinum-based chemotherapies (such as cisplatin) and poly (ADP-ribose) polymerase (PARP) inhibitors (such as olaparib and niraparib). This therapeutic strategy is particularly relevant for triple-negative breast cancer (TNBC) and BRCA1/2-mutant breast cancers, where enhancing the DNA damage response vulnerability can improve treatment outcomes.
