This combination therapy is an investigational regimen being studied by the University of Roma La Sapienza for the prevention of preterm birth in women with a short cervix. It consists of a daily oral supplement containing magnesium (450 mg), palmitoylethanolamide (200 mg), high-molecular-weight hyaluronic acid (200 mg), vitamin B6 (2.6 mg), and vitamin D (2000 IU), administered alongside the standard-of-care vaginal progesterone (200 mg). The treatment aims to reduce the incidence of preterm delivery by leveraging the synergistic effects of its components: progesterone maintains uterine quiescence; magnesium acts as a tocolytic; palmitoylethanolamide (PEA) provides anti-inflammatory effects via PPAR-alpha activation; high-molecular-weight hyaluronic acid (HMWHA) offers structural support and anti-inflammatory properties; and vitamins B6 and D support maternal-fetal immune and metabolic health.
