Magnesium L-threonate is a mineral supplement composed of magnesium and the organic acid L-threonic acid. It is primarily used to prevent and treat low levels of magnesium in the blood. Unlike other forms of supplemental magnesium, it can cross the blood-brain barrier, potentially increasing brain magnesium levels more effectively. This property has led to its investigation for cognitive enhancement, memory improvement, anxiety reduction, sleep regulation, pain relief (including in cancer patients), and neuroprotection. The mechanism involves providing bioavailable magnesium ions that serve as cofactors for over 300 enzymatic reactions essential for bone and muscle function, protein synthesis, nerve transmission, energy production (ATP formation), synaptic plasticity in neurons (which may support learning and memory), and more[1][3][4][5][7]. Magnesium L-threonate was first developed around 2010 as a novel form of supplemental magnesium with improved central nervous system penetration[5]. While animal studies show promise for cognitive benefits and some small human trials suggest improvements in memory or pain management[4], robust clinical evidence remains limited.
