Overview

The Magnitude Group refers to a specific dose-reduction strategy for buprenorphine evaluated in a Phase 2 clinical trial (NCT03107130) led by Steve N. Caritis, MD. This regimen is designed for pregnant women undergoing opioid substitution therapy for opioid dependence. In this strategy, buprenorphine doses are reduced by alternating between 1 mg and 2 mg weekly until the patient either discontinues the medication or reaches the lowest tolerable dose. The active pharmacological agent, buprenorphine, is a partial agonist at the mu-opioid receptor and an antagonist at the kappa-opioid receptor. The study aims to determine how the magnitude of dose reduction impacts maternal withdrawal symptoms, measured via a visual analog scale, as well as neonatal outcomes such as Neonatal Abstinence Syndrome (NAS) scores and neurodevelopmental health.
